Calcium Phosphate Transfection Kit

for introducing genes into cell culture. This product is designed to reduce cell toxicity caused by calcium phosphate salts
compared to conventional methods, making it optimized for transfection in primary cultured neurons where transfection is
considered difficult.

Protocol
Application 1: Evaluation of cytotoxicity after transfection into primary cultured neurons
The product was applied to primary cultured neurons derived from the cortex of fetal mouse brain, and cytotoxicity was evaluated on 10th of culture (3rd day after transfection).
Transfection with this product results in a higher survival rate in primary cultured neurons compared to transfection with conventional products. This result suggests that this product reduces the cytotoxicity caused by calcium phosphate salts.
Application 2: Transfection of pmCherry-N1 vector into primary cultured neurons
The pmCherry-N1 vector was introduced into primary cultured neurons derived from the mouse fetal brain hippocampus using this product, and their subsequent progress was observed.
On the 3rd day after transfection, primary cultured neurons were fixed and observed by fluorescence microscopy. As a result, strong fluorescence of mCherry was observed in the cell body and dendrites, confirming gene transfer into primary cultured neurons.
On the 7th day after transfection, primary cultured neurons were observed using a fluorescence microscope, and the survival of primary cultured neurons emitting mCherry fluorescence was confirmed.
Application 3: Transfection of pAcGFP-N1 vector into Hela cells
pAcGFP-N1 vector was introduced into HeLa cells using this product. The cells were observed using a fluorescence microscope and analyzed using flow cytometry on the 2nd day after transfection.
The result shows that this product also performs effectively on cell line.
