Is that cell toxicity assay accurate?
Results of cell number reduction by cell count reagent such as MTT or WST-8 assay do not show us if cells are dead or simply are not reproducing. However, because LDH cytotoxicity assay enables determination of cell death number, using cell count reagent along with cell toxicity assay more accurately determines on cell toxicity assay. It is not necessary to set up a special assay system; an absorption spectrometer is sufficient when using our cell count reagent kit and LDH cytotoxicity assay kit, which is a superior assay with low cost.

Basics
This product detects free-LDH from cells with damaged cell membranes.

| 1. Dead cells with damaged cell membranes release LDH to cell culture medium. | 2. Free-LDH in medium catalyzes the above reaction, then produces formazan, which has absorption with 490 nm. |
Usage

When using with cell proliferation assay kit, pipet 100 ul of cell suspension up at first, then conduct cytotoxicity assay in another micro-well plate. Cells in the original micro-well plate can be used for cell proliferation assay.
Application
Cell damage assay with melittin
Add represented concentration of melittin to 24-hour-cultured B16/BL6 cell (1 x 104 cells/well), then assay after 30-minute incubation. Substrate solution was directly added to each micro well. The higher concentration of melittin added, the stronger cell toxicity observed.
